Ethidiummonoazidebromide(EMA)isafluorescentnucleicacidstainwithaphotoaffinitylabel.Thedye,afterphotolysis,bindscovalentlytonucleicacids.ThedyehasbeenusedtofootprintdrugbindingsitesonDNA²,tomodifyplasmidDNA,^3,4andforlabelingyeastcellsforleukocytephagocytosisassays⁵.Aparticularlyusefulapplicationofthedyeistolabeldeadcellsinamixedpopulationofliveanddeadcells.BecauseEMA isrelativelyimpermeanttolivecells,itselectivelybindsDNAindeadcells.PhotolysisrendersthedeadcellDNAcovalentlylabeledwiththedye.Onecanthenwashandfixthecellpreparationforanalysisbymicroscopy,fluorescenceplatereaderorflowcytometry.Themajoradvantageofthismethodisthatresearcherscanavoidextensivemanipulationoflivepathogenicorganisms.⁷
