PMA(propidiummonoazide)isahighaffinityphotoreactiveDNAbindingdyeinventedbyscientistsatBiotiumforviabilityPCRofbacteriaandotherorganisms.Thedyeisweaklyfluorescentbyitselfbutbecomesmorefluorescentafterbindingtonucleicacids.ItpreferentiallybindstodsDNAwithhighaffinity.Uponphotolysis,thedyecovalentlyreactswithDNA,thusresultinginpermanentDNAmodification.Thedyeiscellmembrane-impermeable,andthuscanbeselectivelyusedtomodifyonlyexposedDNAfromdeadcellswhileleavingDNAfromviablecellsintact.Thisfeaturemakesthedyehighlyusefulintheselectivedetectionofviablepathogeniccellsbyquantitativereal-timePCRinthepresencedeadcellswhoseDNAhasbeenPMA-modifiedandthuscannotbeamplified.SinceBiotiumfirstdevelopedPMAdye,therehavebeennumerouspublicationsontheuseofthedyeinpathogenicbacterialdetectionrelatedtofoodandwatersafety,medicaldiagnosisandbiodefense;downloadthePMAReferenceList.PMAdyeisalsoavailableinH2O(40019),aconvenientready-to-useformat.
